Futurity.org ? Switching off zinc may stop breast cancer

"Our better understanding of how exactly zinc is delivered suggests if we can block malfunctioning transporter channels, we can potentially halt the growth of these forms of cancer. We believe this makes zinc, and zinc delivery, a high priority for future cancer research," says researcher Kathryn Taylor. (Credit: dream designs / Shutterstock)

CARDIFF U. (UK) ? The body?s control mechanisms for delivering zinc to cells could be key to improving treatment for some types of aggressive breast cancer.

New research by Cardiff University and King?s College London has identified the switch that releases zinc into cells, with important implications for a number of diseases.

Zinc has long been known to play a vital part in human health. Too much zinc, or too little, can cause cell death. A growing body of evidence links zinc to disease states including neurodegeneration, inflammation, diabetes, and cancer.

Zinc levels in cells are controlled by protein molecules called zinc transporters. These move zinc in and out of the cell to ensure correct levels are maintained. Until now, scientists have not understood how the transporters release the zinc.

The research team has identified a switch, known as CK2, a protein that opens one transporter, called ZIP7, and allows the zinc to flow.

Earlier research by the team has already linked zinc delivery to types of breast cancer. Higher levels of intracellular zinc and the ZIP7 transporter were found in tamoxifen-resistant breast cancers. CK2 was also known to be more common in cancers which encourage cell growth.

As reported in the journal Science Signaling, the discovery that CK2 opens ZIP7 suggests that drugs that block this release of zinc could also block cancer development. Early results from clinical trials of CK2 inhibitors suggest they are performing well.

?We know that zinc, in the right quantities, is vital for development, our immune systems, and many other aspects of human health,? says Kathryn Taylor of Cardiff?s School of Pharmacy and Pharmaceutical Sciences. ?But when something goes wrong with the body?s zinc delivery system, it looks as though disease can result. In particular, our research has shown a link to highly aggressive forms of breast cancer.

?Our better understanding of how exactly zinc is delivered suggests if we can block malfunctioning transporter channels, we can potentially halt the growth of these forms of cancer. We believe this makes zinc, and zinc delivery, a high priority for future cancer research.?

Professor Christer Hogstrand from the Diabetes and Nutritional Sciences Division at King?s College London, says: ?Our discovery provides a mechanistic explanation for how the cell uses zinc to regulate different functions.

?The evidence that zinc is released on command in the cell and then regulates cellular processes seems to set it apart from other transition metals, such as copper and iron, in the way that it is used by the body. These findings should open the door for new research into the roles of zinc in health and disease.?

The research is funded by a Wellcome Trust University Research Award.

Online gaming firm 888's full-year revenue up 26 percent (Reuters)

(Reuters) ? British online gaming company 888 Holdings saw its full-year revenue grow 26 percent as the number of customers gambling on its websites grew, and the company said strong current trading continues into the first quarter.

Cash-strapped Britons are staying at home more, watching TV and gambling online, updates from consumer-facing companies showed, as fears grow that the UK will slip back into recession.

The company, which operates 888 casino, poker, sport and bingo brands, posted a revenue of $331 million, up from $262 million a year ago.

As of December 31, the company said it had 10.6 million casino, poker and sport real money registered customer accounts, up 22 percent from a year ago.

The prospects for online betting companies are growing brighter as they seek to break up national gaming monopolies in a number of EU countries and the United States considers legalizing online poker.

Last week, a unit of private equity-owned Caesars Entertainment Corp extended its UK software licensing agreement with 888 to the United States.

Last October, 888 rival Bwin.party Digital Entertainment said it planned to partner with U.S. casino operators MGM Resorts International and Boyd Gaming to offer internet gambling, should it be legalized, to American customers.

888 shares, which have more than doubled from their 52-week low last year, closed at 57.25 pence on Monday on the London Stock Exchange. (Reporting by Jochelle Mendonca in Bangalore; Editing by Joyjeet Das)

Russia pushes Syria reforms as bloodshed mounts (AP)

BEIRUT ? Days after blocking a U.S.-backed peace plan at the U.N., senior Russian officials pushed for reforms Tuesday during an emergency meeting with Syrian President Bashar Assad, promoting a settlement to end the uprising without removing him from power.

Thousands of flag-waving government supporters cheered the Russians in the Syrian capital of Damascus, while to the north, Assad's forces pounded the opposition city of Homs ? underscoring the sharp divisions propelling the country toward civil war.

The violence has led to the most severe international isolation in more than four decades of Assad family rule, with country after country calling home their envoys.

France, Italy, Spain and Belgium pulled their ambassadors from Damascus, as did six Gulf nations, including Saudi Arabia. Germany, whose envoy left the country this month, said he would not be replaced. The moves came a day after the U.S. closed its embassy in Syria and Britain recalled its ambassador.

Turkey, once a strong Assad supporter and now one of his most vocal critics, added its voice to the international condemnation, with Prime Minister Recep Tayyip Erdogan saying his country cannot remain silent about massacres in Syria. He said Turkey would "launch a new initiative with countries that stand by the Syrian people instead of the regime."

His comments reflect a growing movement by the U.S., Europe and countries in the region to organize a coalition of nations to back Syria's opposition, though what kind of support remains unclear. Over the weekend, U.S. Secretary of State Hillary Rodham Clinton called for "friends of democratic Syria" to unite and rally against Assad's regime.

Russian Foreign Minister Sergey Lavrov flew into Damascus on Tuesday, accompanied by his foreign security chief, to try to boost a plan that would keep Assad in power, even though many prominent members of the opposition reject that entirely.

"It's clear that efforts to stop the violence should be accompanied by the beginning of dialogue among the political forces," Lavrov said, according to the Russian news agency ITAR-Tass. "Today we received confirmation of the readiness of the president of Syria for this work."

The visit was also a sign that Moscow wanted to get a firsthand assessment of the situation on the ground in Syria ? and the raucous welcome the diplomats received from thousands of regime supporters appeared aimed at showing that Assad's grip is firm, at least in Damascus.

Syria has been a key Russian ally since Soviet times, and Moscow remains a major arms supplier to Damascus even as Assad unleashes his forces to crush not only peaceful protesters, but army defectors who are fighting the regime.

The U.N. estimates the government crackdown has killed more than 5,400 people since March, making Syria's conflict one of the deadliest of the Arab Spring. Hundreds more are believed to have died since the U.N. released that figure in January, but the chaos in the country has made it impossible for the world body to update its figures.

Tuesday's visit by Lavrov and intelligence chief Mikhail Fradkov was evidence that Russia does not want to be seen as giving Assad a free hand to crush his opponents in the wake of Saturday's veto at the U.N. Security Council.

Both Russia and China blocked a Western- and Arab-backed resolution supporting calls for Assad to hand over some powers as a way to defuse the 11-month-old crisis.

Russia has opposed any U.N. call that could be interpreted as advocating military intervention or regime change. Russia and China also used their veto powers in October to block an attempt to condemn the violence in Syria.

On Tuesday, Moscow delivered its own message to Syria, calling on all sides to hold a meaningful dialogue.

"Necessary reforms must be implemented in order to address legitimate demands of the people striving for a better life," Lavrov told Assad, according to ITAR-Tass."

Assad replied that Syria is determined to hold a national dialogue with the opposition and independent figures, saying his government was "ready to cooperate with any effort that boosts stability in Syria," according to the Syrian state news agency SANA.

SANA also said a committee tasked with drawing up a new constitution has finished work and will submit its recommendations to the president. The panel said earlier that it would recommend abolishing Article 8, which states that the ruling Baath Party is the leader of the state and society.

The article's abolishment was once a key demand of the protest movement. However, such overtures are now unlikely to satisfy opposition leaders who say they will accept nothing less than the downfall of the regime.

Repeated efforts by the Arab League and Russia to broker talks have been rejected by the Syrian opposition, which refuses any negotiations while the crackdown continues. The opposition has also said Assad's proposed reforms, including a new constitution and eventual multiparty elections, are aimed at keeping his hold on power.

In Tuesday's talks, Assad told Lavrov that Russia's position has played "a key role in saving our motherland," according to ITAR-Tass.

As Lavrov's convoy snaked its way along Damascus' Mazzeh Boulevard, it was greeted by a sea of Assad supporters cheering the vetoes at the U.N.

"Thank you Russia and China," read one banner that had photos of Assad and the Russian president. Many stood in the rain carrying Syrian flags as well as the red, blue and white Russian banner.

"I am here to thank Russia for its stand in the face of the world conspiracy against Syria," said Manya Abbad, 45, as she waited for Lavrov's convoy. "I wish the Arabs adopted similar stances."

The Assad regime says terrorists acting out a foreign conspiracy to destabilize the country are behind the uprising, not people seeking to transform the authoritarian regime.

But in the flashpoints of the conflict, witnesses, residents and human rights workers say Assad's forces are shelling and firing indiscriminately. On Tuesday, the troops renewed their assault on one of the main centers of the opposition, the city of Homs, with activists saying tanks were closing in on a restive neighborhood.

Despite the assault, members of the rebel Free Syrian Army pledged to protect residents of the besieged Baba Amr neighborhood.

"We are just here to respond and defend the local residents from Assad's army snipers," said one fighter, according to Associated Press television footage.

Shielded in the corridors of a deserted building once occupied by Assad's forces, the rebels moved carefully from one position to another overlooking suspected sniper hide outs.

At a makeshift medical clinic, the dead were wrapped in white sheets and piled on a pickup truck outside. Doctors appeared overwhelmed by the number of wounded and the severity of their injuries.

"Can someone help, please!" wailed a man kneeling by a wounded relative on the floor, "Someone come and see him!"

Homs was the site of the deadliest assault of the uprising on Saturday, when activists reported more than 200 people were killed in an overnight bombardment hours before the U.N. vote. The government denied the deaths.

Syria has blocked access to trouble spots and prevented most independent reporting, making it nearly impossible to verify accounts from either side.

In Washington, meanwhile, the Obama administration said the U.S. was not considering arming opposition groups in Syria amid calls from some U.S. lawmakers to consider such an option.

White House spokesman Jay Carney said that at this point, U.S. efforts were focused on exploring the possibility of providing humanitarian aid to the Syrian people and ratcheting up the pressure on the Syrian government.

U.S. senators John McCain and Joe Lieberman spoke forcefully Tuesday in favor of arming the opposition.

"It's an option that now should be on the table," McCain said. "The situation has deteriorated to where we need to consider every option, including no travel zones, including sanctuaries, including getting the Turks to help the Syrian nationals."

McCain conceded the situation in Syria was more complicated than in Libya, where opponents of Moammar Gadhafi quickly gained control of an eastern city, but insisted it was necessary.

"I feel very strongly that what's happening in Syria is exactly what we got into Libya to stop Gadhafi from doing. And Assad is doing it to his people," said Lieberman. "The question is what do you do. One of the things is giving support to the Syrian Free Army."

America is becoming a nation of renters

By John Schoen

?

There was fresh data from the government Tuesday showing that the American dream?of owning a home is fading fast.

The share of all U.S. privately-owned houses that stood empty?fell in the fourth quarter to its lowest since 2006 as the number of houses occupied by renters rose faster than the pace of new vacancies created by homeowners moved out, according to the Commerce Department. ?There?number of housing units occupied by renters rose by 749,000 in the fourth quarter?compared?to a year earlier; some 91,000 fewer homes were occupied by owners, the data show.?

With the fast pace?of foreclosures showing no sign of letting up, the U.S. homeownership rate continues to fall. Just 66.0 percent of U.S. homes were occupied by their owners in the fourth quarter of last year ? half-a-percentage-point lower than a year earlier. That?s the lowest level of homeownership since the second quarter 1998.

The steady rise in demand for rentals has tightened the supply of housing, helping to push rents higher. Homebuilders report that the market for multi-family units has been a lone bright spot in the overall housing market. While the pace of new household formations has lagged historical averages, those new households are becoming renters. That trend is likely to continue until would-be home buyers gain more confidence?that the housing market has bottomed, according to NAHB Chief Economist David Crowe.?

Despite the ongoing high rate of default and foreclosure, the share of houses that stood empty fell in the last three months of last year. Part of the reason may be that bankers have been trying to match the pace of new foreclosures to the sale of homes they?ve already seized, waiting to sell a house before closing out the foreclosure of another one. By letting homeowners remain, banks can reduce the cost of maintaining a house until they?re ready to put it on the market.

Though the overall share of?houses that stood empty fell slightly at the end of last year, it?remains high by historical standards. A separate report?by the Government Accountability Office?in November, based on Census data, found that the number of vacant, non-seasonal, residential properties?increased 51 percent nationally,?from nearly 7 million in 2000 to 10 million in April 2010.

The homeownership rate will likely continue to fall until the?pace of foreclosures begins to ease. So far, public and private efforts to modify loans to allow homeowners to stay put haven?t kept up with new defaults. With home buying demand weak, those newly-vacated homes will continue to weigh on home prices.

By the end of the third quarter of last year, some 12.6 percent of homeowners with mortgages ? or more than 6 million homeowners - were either delinquent on their payments or in foreclosure, according to the Mortgage Bankers Association. Roughly 22 percent of residential properties with mortgages were underwater at the end of the third quarter, according to CoreLogic.

?Add to this the currently high unemployment and underemployment rates, one gets a recipe for further price declines,? said Patrick Newport, an economist at IHS Global Insight. ?Our view is that foreclosures, excess supply, and weak demand will drive prices down another 5 to 10 percent.??

Egypt army fires general to boost image (Reuters)

CAIRO (Reuters) ? Egypt's military leader has sacked the general responsible for media affairs to bolster an image tarnished by killings of protesters and accusations that the men in uniform are undermining Egypt's democratic revolution.

The change is the first in the military council since the generals took power from President Hosni Mubarak during a popular uprising last February.

Although it defused a violent confrontation by ushering Mubarak out, the military has also tried to crush subsequent protests by force, killing dozens. It has only grudgingly agreed to hand over to a civilian president by June, and tried to protect its privileges and avoid civilian oversight.

Major General Ismail Etman, 60, was "exempted from service and replaced by Major General Ahmed Abu El-Dahab, the director of the artillery division," a defense ministry source said. The decision was announced later by state media.

Since the 1973 war against Israel, Egypt's army was seen as a respected institution set above the fray. But its direct involvement in politics has exposed it to closer scrutiny.

The generals are not trusted by many young pro-democracy campaigners, who suspect they want to curtail civilian power by exploiting the fragile security situation.

Dozens died when the army tried to suppress protests on the streets of Cairo in November and December and video of soldiers mistreating injured demonstrators sparked widespread anger. The army said troops were also killed.

It has blamed the violence on "invisible hands" determined to sow chaos among Egyptians and undermine the achievements of the uprising against Mubarak.

"NEW LEADERSHIP"

A source close to the military council said it was concerned about its deteriorating public image.

"The Supreme Council of the Armed Forces concluded that the army's image in the media has suffered over the past year under Etman's leadership," said the source.

"The decision is intended to bring in new leadership to improve the armed forces' performance," and was decreed by Field Marshal Mohamed Hussein Tantawi, who heads the council, the source added.

Etman led the army department which handled a wide range of issues ranging from media relations to dealing with the concerns of military officers.

Little is known about Etman's successor, Abu El-Dahab, and it remains to be seen whether the change will alter the army's public affairs policy.

Tantawi has tried to improve the military's public image, calling on Egyptians to unite with the army and ordering the formation of a committee of generals to ensure positive media coverage, Egyptian media reported last week.

While no longer a member of the 20-member military council, Etman will however remain one of Tantawi's many advisers, positions given to officers closely tied to the army leadership, the source at the defense ministry said.
